Plant Regeneration and Flavonoid 3¢¥,5¢¥-Hydroxylase gene Transformation of Dendranthema zawadskii and Dendranthema indicum

Abstract

|
A method has been developed to regenerate shoots directly from internodes and leaf pieces of the Dendranthema indicum and Dendranthema zawadskii. Optimal shoot regeneration was obtained in BM medium supplemented with BA 3§·/L and IAA 0.2§·/L. In this condition, transgenic plants were generated using the disarmed strain of Agrobacterium tumefaciens LBA 4404, carrying flavonoid 3¢¥,5¢¥-hydroxylase gene. Chimeric flavonoid 3¢¥,5¢¥-hydroxylase (F3¢¥,5¢¥H) construct was prepared by inserting F3¢¥,5¢¥H cDNA on the downstream of CaMV (Cauliflower Mosaic Virus) 35S promoter of pBI 121. PCR and Southern analysis of shoots that were able to root on kanamycin confirmed the presence of the npt II gene in the plant genome.
